An alkaline phosphatase was partly purified from amphioxus by extraction, ammonium sulfate fractionation,pH regulation and get filtration with Sephadex G-100.Preliminary kinetic studies of the enzyme were undertaken. The results are as follows:The optimum pH of alkaline phosphatase for the hydrolysis of disodium phenyl phosphate is 10.1,and the optimum temperature is 40℃ .The Michaelis -Menten constant Km is1.13×10-3M at pH 10.1.Under given experimental conditions,the Michaelis-Menten constant Km varies with different pH values whereas the maximal velocity Vmax remains unchanged which indicates thatthe effect caused by pH is of the competitive type.The alkaline phosphate is strongly activated by Mg++ ion hut is inhibited by Zn++, EDTA,DFP,2-mercaptoethanol and PCMB. The Ki's of three inhibitors were determined and were found to be of the noncompetitive type. The marked inhibition of 2-mercaptoethanol on the enzyme suggests that the disulfide bond is easential to the maintainance of the enzyme activity. From the v versus pH plot,the pK value of the dissociation of the active group is 9.82 which suggests that -NH2 group might be an active group of the enzyme.The energy of activation is 3.16 kcal/mole, the temperature coefficient Q10(at 30-40℃)is 1.15.
从厦门文昌鱼中部份提纯一种碱性磷酸酶,并对该酶作用的动力学进行了初步的研究。该酶作用于底物磷酸苯二钠的最适pH为10.1,最适温度为40℃,测得其米氏常数值(K_m)为1.1×10~(-3)M,pH影响K_m值而不影响最大反应速度(V),表现为竞争性类型。Mg~(++)有明显的激活作用,而Zn~(++),EDTA,DFP,ME及PCMB等在一定浓度范围内,表现为非竞争抑制类型。ME的抑制作用表明硫硫键是维持酶活力所必需的。从pH对酶的活力影响,测得有关酶活性基团解离的pK值为9.82,在不同温度条件下,测得该酶的活化能为3.16仟卡/克分子,其温度系数为1.15(30—40℃)。
Three Toxic components, designated Aa-Toxin Ⅰ,Ⅱ,Ⅲ were isolated from the venom of Agkistrodon acutus by DEAE-sephadex A-50 chromatography and were further purified by gel filteration on Sephadex G-73, and by column chromotography on DE52 cellulose and Cm-sephaqex C-25. All these components show single band on acrylamide gel electrophoresis and one precipitin line on immuno-electrophoresis. They are distinct from each other immunochemically. Aa-Toxin Ⅰ and Aa-Toxin Ⅱ are acidic proteins, Aa-Toxin Ⅲ is basic. The Toxins demonstrated in our work have similar tool. wt about 22000. In addition to hemorrhagic and lethal activities, all these Toxins possess caseinolytic activities. The hemorrhagic and caseinolytic activities of these Toxins are inhibited by EDTA,cystein and the serum of homologous snake. disulfide bond and metals is probably involved in the stabilization of the conformation of these proteins. The amino acid composition of the Aa-Toxin Ⅰ was determined.

The human hair keratin was examined by means of X-ray photoelectron spectro-scopy(XPS).The oxidation of disulfide to sulfonate is the major transformation whenthe hair is treated with hydrogen peroxide,but there have been much controversyin literatures about it.In addition,the oxidation of disulfide bonds at different positionson the surface of human hair has been revealed by XPS for the first time,approximately50% of the disulfide bonds in surface of long hair tip ends are oxidized duringweathering,and 20 to 30% of the disulfide bonds in surface of moderate long hair areoxidized during weathering.The mechanisms for oxidative degradation of disulfidesare discussed.A suggestion that this reaction is occurring largely by S-S fission,butcannot entirely rule out the possibility of C-S fission.
本文用电子能谱验证了头发角蛋白被H_2O_2等氧化剂氧化时二硫键的主要氧化产物是磺酸基团,从而回答了文献上有关这个问题长期存在的争论。此外文中首次用电子能谱揭示了长发不同部分的表面二硫键被自然氧化的不同情形,如揭示了长发末梢表面竟有50%左右的二硫键被自然氧化,中长发表面也有20~30%的二硫键被自然氧化。本文最后探讨了二硫键氧化降解机理,认为S-S 氧化断裂是主要的,C-S 断裂的可能性也不能完全排除。
